A winner is someone who never cheats or gives up, but can also accept defeat when all other means have been exhausted. Even if they can not reach their goal, they tried every way that they could while keeping their honor.
A loser/failure will either cheat to reach their goal or give up without even trying.
What do they win or lose? Self respect and the respect of others.

Sometime winner can be used ambiguously like "your a real winner". Then a rock star can yell from the stage and say, "attention all you loser". A winner shows 101% while a loser shows 98%. what ever.

A winner is someone who always tries, and who never gives up on themselves or their goals.
A loser/failure is someone who never tries in the first place.

I think it's an attitude. Losers are usually passive and think that stuff just 'happens'; winners go out and knock on doors until somebody answers.

Depends on the situation.
Winning may be a mindset or an actual label. Same goes for losing.

Charlie Sheen is a winner.
My cousin is a loser, he didn't go to college and has like 5 kids with a crackwhore.
